Transport of methotrexate by the choroid plexus.

چکیده
Methotrexate-3H was accumulated in vitro against a con centration gradient by the rabbit choroid plexus. This accumu lation was energy dependent, inhibited by metabolic inhibitors, and exhibited self-saturation and competitive inhibition. Several folie acid analogs with a 4-amino group were inhibitors of this accumulation, while folie and folinic acid, compounds which do not contain a 4-amino group and are not effective antimetabolites, were ineffective as inhibitors of Methotrexate-3H accu mulation. When Methotrexate-3H clearance exceeded inulin clearance, no accumulation of Methotrexate-3H was demonstrable in vivo. This suggests that in vitro accumulation is equivalent to in vivo transport from cerebrospinal fluid to blood. Such a sys tem would influence the spinal fluid levels of orally or parenterally administered Methotrexate, as well as the systemic toxicity from intrathecally administered drug.

Abstract Background: Choroid plexus (CP) is a branched structure made up of a single layer of epithelial cells and blood capillaries, forming the blood-CSF-barrier. The CSF (cerebrospinal fluid) is mainly produced from the CP. Aquaporin1 (AQP1), water channels that are highly expressed on the apical side of the membrane in choroid plexus, have a major role in mediating water transport across th...

One function of the vertebrate choroid plexus (CP) is removal of potentially toxic metabolites and xenobiotics from cerebrospinal fluid (CSF) to blood for subsequent excretion in urine and bile. We have used confocal microscopy and quantitative image analysis to follow transport of the large organic anion fluorescein-methotrexate (FL-MTX) from bath (CSF side) to blood vessels in intact rat CP a...

Background and Aim: Intracranial physiologic calcification is not accompanied by any evidence of disease and has no demonstrable pathological cause. It is often due to calcium deposition in the blood vessels of different structures of the brain. Computed tomography (CT) is the most sensitive means of detection of for this calcification. The aim of this study was the assessment of the intracrani...

CPCs are a relatively common finding during the second trimester sonography. when a choroid plexus cyst is identified, the presence of structural malformation and other sonographic markers of aneuploidy should be assessed with a detailed fetal anatomic survey performed by an experienced person. if no other sonographic abnormalities are present, the CPC is considered isolated. given the consider...
